Great Britain The Nucleotide Sequences Recognized by Endonucleases Aval and A vail from Anabaena variabilis

چکیده
This sequence is cut as shown by the arrows to give single-stranded 5'-tetranucleotide protrusions (cohesive ends). Endonucleases SmaI, XhoI and XmaI recognize different symmetrical subsets of this sequence and provide independent evidence for the occurrence of these subsets at particular endonuclease-Aval cleavage sites in the bacteriophage-A genome. Further evidence for this structure came from the demonstration that DNA fragments generated by endonuclease AvaI can be ligated to form a discrete set of larger molecules and from nearest-neighbour analysis which showed that cytosine residues occurred at the 3'-side of cleavage points. The observation that endonuclease AvaII recognized a subset of the sites recognized by AsuI [Hughes, Bruce & Murray (1979) Biochem. J. 185, 59-63] led to the deduction that AvaIl recognizes the pentanucleotide sequence:

Transcriptional and mutational analysis of the uptake hydrogenase of the filamentous cyanobacterium Anabaena variabilis ATCC 29413.
A 10-kb DNA region of the cyanobacterium Anabaena variabilis ATCC 29413 containing the structural genes of the uptake hydrogenase (hupSL) was cloned and sequenced. In contrast to the hupL gene of Anabaena sp. strain PCC 7120, which is interrupted by a 10.5-kb DNA fragment in vegetative cells, there is no programmed rearrangement within the hupL gene during the heterocyst differentiation of A. v...
متن کاملMolecular Analysis of the psaC Gene Encoding the F(A)/F(B) Apoprotein of Photosystem I in the Filamentous Cyanobacterium Anabaena variabilis ATCC 29413.
In cyanobacteria, green algae, and higher plants, the psaC gene encodes PsaC, the apoprotein for two iron-sulfur clusters, FA and FB, located in the PSI complex in the thylakoid membrane. FA and FB act as the terminal electron acceptors in the PSI complex (reviewed in ref. 2). Our research objective is to develop a genetic system in which various hypotheses concerning the structural and functio...
متن کاملCharacterization of genes for an alternative nitrogenase in the cyanobacterium Anabaena variabilis.
Anabaena variabilis ATCC 29413 is a heterotrophic, nitrogen-fixing cyanobacterium that has been reported to fix nitrogen and reduce acetylene to ethane in the absence of molybdenum. DNA from this strain hybridized well at low stringency to the nitrogenase 2 (vnfDGK) genes of Azotobacter vinelandii. The hybridizing region was cloned from a lambda EMBL3 genomic library of A. variabilis, mapped, a...
متن کامل5-Aminolevulinate Synthesis in Permeabilized Filaments of the Blue-Green Alga Anabaena variabilis.
Filaments of the blue-green alga Anabaena variabilis permeabilized by dimethylsulfoxide (DMSO) produce increased amounts of 5-aminolevulinate in the presence of levulinic acid. The metabolic activity of the filaments remains unperturbed in the presence of up to 7.5% (v/v) DMSO. Studies utilizing DMSO-permeabilized filaments confirm that 5-aminolevulinate is synthesized preferably from glutamate...
متن کاملCharacterization of genes for a second Mo-dependent nitrogenase in the cyanobacterium Anabaena variabilis.
Anabaena variabilis ATCC 29413 is a filamentous heterocystous cyanobacterium that fixes nitrogen under a variety of environmental conditions. Under aerobic growth conditions, nitrogen fixation depends upon differentiation of heterocysts and expression of either a Mo-dependent nitrogenase or a V-dependent nitrogenase in those specialized cells. Under anaerobic conditions, a second Mo-dependent n...
